Enzymatic Repair of Oxidative DNA Damage1

چکیده
Oxidative DNA damage, including both mutamente and cytotoxic le sions, is implicated in aging and cancer. Studies of the processes which correct such damage in mammalian cells are, however, still in their early stages. Here we have summarized our recent work which demonstrates new features of mammalian oxidative UNA damage repair, such as (a) a functional role for polylADP-ribosyllation in the rejoining of UNA strand breaks and (b) the detective repair of oxidative DNA damage in xeroderma pigmentosum cells.

OGG1 DNA Repair Gene Polymorphism As a Biomarker of Oxidative and Genotoxic DNA Damage
Background: Single nucleotide polymorphisms in 8-oxoguanine DNA glycosylase-1 (OGG1) gene modulates DNA repair capacity and functions as one of the first lines of protective mechanisms against 8-hydroxy-2’-deoxyguanosine (8-OHdG) mutagenicity. OGG1-Cys326 gene polymorphism may decrease DNA repair function, causing oxidative stress due to higher oxidative DNA damage. The main purpose of this stu...
متن کاملMitochondrial DNA repair and association with aging--an update.
Mitochondrial DNA is constantly exposed to oxidative injury. Due to its location close to the main site of reactive oxygen species, the inner mitochondrial membrane, mtDNA is more susceptible than nuclear DNA to oxidative damage. The accumulation of DNA damage is thought to play a critical role in the aging process and to be particularly deleterious in post-mitotic cells. Thus, DNA repair is an...
متن کاملAdvances in Carcinogenic Metal Toxicity and Potential Molecular Markers
Metal compounds such as arsenic, cadmium, chromium, cobalt, lead, mercury, and nickel are classified as carcinogens affecting human health through occupational and environmental exposure. However, the underlying mechanisms involved in tumor formation are not well clarified. Interference of metal homeostasis may result in oxidative stress which represents an imbalance between production of free ...
متن کاملStudies on electron beam induced DNA damage and repair kinetics in lymphocytes by alkaline comet assay
Background: Exposure to ionizing radiation is known to induce oxidative stress followed by damage to critical biomolecules like lipids, proteins and DNA through radiolysis of cellular water. Since radiation has been widely used as an important tool in therapy of cancer, the detailed investigation regarding the DNA damage and repair kinetics would help to predict the radiation sensitivity of cel...
متن کاملInfluence of diet on oxidative DNA damage, uracil misincorporation and DNA repair capability.
The contribution of diet to cancer ranges from 10 to 80%. The low ingestion of antioxidants and enzymatic cofactors involved in DNA repair and methylation reactions and the high ingestion of chemical additives present in the modern diet, associated with genetic factors, could lead to genomic instability and the hypomethylation of proto-oncogenes, thus contributing to development of genetic-rela...
